Samuel's curse or poison works like this it destroys the body first and then the soul until there is nothing left .
In some cases if the person has high demonic powers they will with stand it for a while but will succumb to the curse/poison.
Vail with stand it for a while because of his inherited high demonic powers.
Issei verl little to no demonic powers so could not fight with stand it .
For Ophis I think that she was too strong to have gotten damage. All Samuel did was restraint and steal her power.
If I remember right in vol 9 the Hero faction tried to summon GR to See what kind of effect Samuel would have on him.
There is no way to know what kind of effect Samuel would have on him and sense Ophis is like him a Divine Dragon there also no way to know if it can kill her or damage her soul.
