There's some pretty huge differences between Tsukune and Yuusuke actually. For starters, Yuusuke already had the demon blood when he was born, but he had to die in order for it to activate/awaken because his body worked like a human's. When he died, it allowed the core of his demon energy to take its place and this is what transformed him into a demon. Kurama told Yuusuke that demon hearts don't work like human hearts so it wouldn't beat the way a human heart does, which is why death for Yuusuke makes sense. Tsukune is entirely different however. He was born human (like Yuusuke) and never had demon blood when he was born (unlike Yuusuke). Death wouldn't help Tsukune in this case at all. The blood he receives from Moka at the beginning of the series only works to transform him into a Vampire for a limited amount of time before losing its potency. This is likely due to the fact that human restorative energy is nothing compared to a Vampire's, so Tsukune's spirit could only use the power for a short time. It's a bit similar to the Talent in "The Nightangel" trilogy where there are individuals who have power, but no ability to restore the power after using it (and are thus only able to use Talent once, maybe twice throughout their life). It is because Tsukune is human that he cannot restore his demon energy. However, while the demon energy in Moka's blood is lost after being used once, Moka's DNA remains, and this is what increased Tsukune's energy restoration and the more blood he took from Moka, the more power was he able to use and eventually store, which lead to him losing his mind and entering into a ghoul-like state.
What the ritual is doing now is turning his body completely into that of an Ayashi i.e. a Vampire. This is bound to increase his restorative power. He'll be able to use the Talent not just once or twice, but anytime he wishes to because his DNA is geared toward restoring this power. The only similarity between Tsukune and Yuusuke would be their inability to properly control the power after immediately transforming into a demon/ayashi because while their body is completely transformed, their memory and consciousness remains human, so their memory of using their power is different. In Yuusuke's case, he was more used to handling Spirit Energy rather than Demon Energy (hence why he missed Sensui with his Spirit Gun in the battle that followed his transformation). Tsukune, however, isn't used to handling Demon Energy or Spirit Energy at all. Again, this is similar to something that happens in "The Nightangel" trilogy where the main character used so much Talent that he lost control of it and literally started glowing. When Tsukune is going through now is similar because his mind is still not used to the pressure of the Demon Energy to his consciousness.
