It seems the story for most of the characters are similar to the game, but I am not sure about Asami's. People say her profile foreshadowed this. Why? There wasn't a bit about how she likes Shuri. Violent, yes. But lesbian? No.

Okay, she's a lesbian in the anime.

But in the game, you have to play a guy which means it will be romance between a heterosexual relationship. Meaning 1 of 3 things:
1. The anime pulled off this s*** to piss people off and rush an ending.
2. In the game, in Asami's path, your goal is to get her to go back to being straight.
3. In the game, in Asami's path, your goal is to get Asami with Shuri.
