The anime introduced different character as the homunculus. It also differed on who and how homunculus are created. Below is a running list of the difference:
Lust: Both anime and manga are the same character. Possess the Ultimate Halbred (her finger can be extended into razor sharp blade that cuts through stones). Roaming around the country with Envy and Gluttony. In the anime, Lust was created by Scar's alchemist brother.
Envy: Both anime and manga are the same character. Shapeshifter. Roaming around the country with Lust and Gluttony. In the anime, it's suggested that Hoenheim Elric created Envy.
Gluttony: Both anime and manga are the same character. Consumes everything. Roaming around the country with Lust and Envy. In the anime, it isn't clear who created Gluttony, though Hoenheim Elric is a strong candidate.
Greed: Both anime and manga are the same character, though in the anime, Greed is imprisoned in the 5th Research Lab for 100 years, while in the manga he roam free on his own for a century. In the anime, Ed killed Greed after being tricked by his creator Dante. In the manga, he is unmaked by Father after being incapacitated by Wraith. In both instance, Greed possess the Ultimate Shield, which allows him to turn his skin into diamond-like surface. Also in both version, Greed rebelled against his creator. In the anime, Dande created Greed.
Sloth: In the anime, she's a homunculus created by Edward and Alphonse. She later adopted the name Julia Douglass. In the manga, Sloth is a hulking man in chain, supposedly forced to work by Father. The anime version of Sloth can turn into water.
Wraith: In the anime, he was the homunculus created by Izumi Curtis. He also grafted Edward's arm and leg. He can subsumed any object. In the manga, King Bradley is Wraith, and he subdue Greed to be unmake by Father. Also note, manga's Wraith seemed to be the only homunculus that can age, thus allowing him to be able to pass as a human.
Pride: Has yet to appear in the manga. In the anime, King Bradley is Pride. In either case, King Bradley of both version possess the Ultimate Eye (under his eye-patch) which some have speculated that he could use to forsee possible outcome in the very near future, thus allowing him to completely anticipate his opponent's movement. He's also very fast.
In the anime, homunculus are the end result of failed human transmutation. In the manga, all of the homunculus are the creation of an semi-unseened alchemist named Father. Father appears to be able to cut off pieces of his own soul and encapsulated it into a small Philosopher's Stone to serve as base of the homunculus. We've seen this with Greed and Lust.
