I would definately say that the "Coolest" gal in manga, in my opinoin at least, would be Maj. Motoko Kusanagi from Masamune Shirow's "Ghost in the Shell."
As for a "coolest" male character, I suppose that would be a lot more difficult. I'd say that "Lone Wolf" Ogami Itto is an interesting character (and total badass), but he's not exactly someone you'd want at a party, neh?
