After all, if blood loss would actually cause Tsukune's mutation to reemerge, then we should have already seen some symptoms of that.
Since, we shouldn't forget that Tsukune was pierced by Alucard as well, and he definitely lost some amount of blood, because of it, before his regeneration ability kicked in.
While, it's possible that this development related with Akua might push Tsukune's transformation to some new stage, considering Tsukune's determinator nature, as well as the things that he survived so far, when it comes to his transformation, he is probably going to survive the next portion of developments as well.
Basically, I don't see Ikeda actually killing off Tsukune, no matter how dark the story seems to be.

Spoiler for Reply:
It would be hard for me to believe that Akuha can not to put 2 n 2 together in just looking at Tsukune's body and energy signature and see that he has some connection to Alucard. Someone who is smart enough to figure out how to perform a perfected Jingentou just by watching can't see that the energy signature of her sister's boyfriend is virtually the same as Kaiju Alucard?
And I'm not saying that Tsukune will sucumb to this either - it is just a possible route.
