uh no..if it's two audio layers..and the instrumental is on one layer..and the vocals are on the other layer..it's very easy to retune the vocals without affecting the instrumentals (since it's on a different layer) and then after doing so, compile it as a single audio file

Vocals can be more or less extractly fairly easily by subtracting a sample-accurate-shifted version of the instrumental from the actual song. Your success depends on the amount of vocal-volume induced dynamic compression applied to the song and also how badly mastered the song was.
