I do not know if they are the most romantic but my favorites are:

1) The Lake of Despair scene in Princess Tutu
2) The hand holding scene in Kare Kano

In the Kodomo no Jikan second season, the way the flowers appear slowly and spread gradually around the couple while they kissed until the flowers cover them is the most romantic moment I've seen. I won't spoil the moment by revealing the couple's names so I am hiding it in the spoilers.

edit: Oops I didn't realize I replied to this thread before, where here is an additional one I did not mention in my last post.

Although I have not seen the anime I also love the scene in the Maison Ikoku manga when Godai says

This scene really showed the growth and maturity of Godai and how his crush on Kyouko had really blossomed into real genuine love.

Interestingly they tried to do something similar in the recent Natsuyuki Rendezvous, although the scene did not work as well for me personally.

I'll go with a more generic choice and choose Kimi ni Todoke's confession scene. You have to give it to KnT, with an entire season of buildup culminating in that one scene full of sweetness and KYAAAA'ing (in my part).

I hate to agree with the Obelisk, but I have to second (er, third), this. Even as much as I hate Minmay, even she has a few good scenes throughout the series where I really felt something for her, though Misa obviously stole the show when it comes to the heartwarming romance side of things. Unlike Obelisk, I very much enjoyed Frontier, but I'm not ashamed to admit that the high point of Macross was the first Macross.

Some other great anime romances:

1) Lafiel and Jinto in the Banner/Crest of the Stars shows.
2) Ray and Charles Beams from Eureka Seven.
3) Nataru and Birdy from Birdy the Mighty Decode:02.
4) Toshihiko and Yuka from Kemonozune. Something I almost missed due to being so turned off by the art style at first.
5) Kenshin and Tomoe from Rurouni Kenshin: Tsuiokuhen.

Strangely enough, while I've enjoyed more than my share of "romance" anime, it's the romances in anime from other genres that I tend to enjoy the most.

To me, most romantic scene would be the everything before the confession. You know, so-called fated or coincidence meeting, then friendship, arguments, love-hate, jealousy... everything that stirs your feelings, until one of the two says "i love you..", then after that it became so obvious, that that feelings became no more, and slowly faded till the end of the series.

But of course this only applies for most original series, not adaptation of novel or manga (that i have read before), or even worst, from VN where you can expect ridiculous or exaggerated romance plot.

But just to follow the thread, the most romantic scenes for me would be

Hmm one I like is where Amu Hinamori From shugo chara Reaches for something and Ikuto falls on top her. I haven't watched much romance anime so I couldn't think of a better scene than this. but it was still so~
