I believe these fit all/most your requirements:
Hoshi wo ou kodomo Spice & Wolf Sakasama no Patema - although the villian is kinda one dimensional Castle in the Sky, Kiki's Delivery Service, Spirited Away and other Studio Ghibli films Kami sama no Inai Nichiyoubi

Akatsuki no Yona is a must here along with Seirei No Morbito.
Inuyasha is another one. Kamisama no Inai Nichiyoubi - it has few issues but over all really nice series that deal with adventure and in a way reincarnations/god and etc. MC is not action based but she is likeable. Beautiful animation and soundtracks. Mai HiME - not adventure based but it is action with magic elements, full of very likeable and memorable female characters who can kick ass. Romeo x Juliet possibly? Anime differs from classic and has magic and everything. Senjou no Valkyria: Gallian Chronicles war based but has some magical elements later on. Soul Eater is another big title. Toaru Kagaku no Railgun - not adventure or magic but characters have super powers like controling electricity , teleportation and etc.

Claymore is a possibility, though the heroine doesn't laugh that often. Some of the antagonists, Riful in particular, are pretty droll though. On Funimation.
The girls in Mahou Shoujotai Arusu (aka "Tweeny Witches") have strong personalities and some powers, but they're all between 10 and 14 if that matters. There are some solid adult characters in supporting roles. Finally I'll suggest Kemono no Sou-ja Erin which follows a young girl from about eight to eighteen. She wants to follow in her mother's footsteps and become a veterinarian to magical beasts. Eventually she becomes the target of competing factions in a civil war. On Crunchyroll.

I haven't seen Akagami no Shirayuke-Hime, but I strongly second Akatsuki no Yona, Twelve Kingdoms, and Seirei no Moribito. Actually, I'd say all three of those series are in my short list of all time favorites. Akatsuki no Yona just aired recently and the manga is still ongoing, it's really good.

I also agree with Serei no Morobito and Twelve kingdom. Anyway:
Arcana Famiglia: La Storia Della Arcana : Not my thing, but not bad and fit the bill Witchblade Blood+/Blood C Yunketsu no Maria Nanoha strikers (it has some prequels though) Kaibutsu Oujo Murder Princess - This one is really fun. Scrapped Princess - Pretty much classic Read or die Wagaya no Oinarisama - it's low budget stuff, but novels got some relevant prizes, it's also my personal pick. Vakyria chronicless If you don't mind sci-fi, these are my favourites Birdy the mighty decode Nobunagun The Third

Kemono no Souja Erin and the two seasons of Saiunkoku Monogatari should definitely be added to the list of:
Twelve Kingdoms Seirei no Moribito Akatsuki no Yona The currently airing Akagami Shirayuki is also an excellent choice. From movies the Ghibli productions are excellent suggestions too. Some of my faves were already mentioned, and include: Only Yesterday Whisper of the Heart Kiki's Delivery Service Princess Mononoke Spirited Away Nausicaa The World Masterpiece Theatre works are also quite good, but have many episodes. SomE series I enjoyed are: Akage no Anne Little Princess Sara Perrine Monogatari

noein and shin sekai yori. two of the best shows ever made probably
both have: 1. strong MC's who aren't serious all the time 2. some of the best side characters/villains. edit: Add revolutionary girl utena to this; a real gem |
