For me it was Yuuko Ichihara from XxXHolic, there was something very magnetic about her personality and wisdom, needless to say i read the whole manga and watched all the seasons plus OVA.

I have to admit, I am realy player. I can't even remember if my first were Motoko Aoyama or Inner Moka (manga only)...

Mine would be Asuka, from the original Evangelion series. I'm sure I'm not alone.
I watched the series a good decade ago (and the fact that it's been that long is sort of scary; time really does fly), and I had never seen anything like her before, especially not animated. Her gorgeous looks, and her spirited and abrasive personality played their parts, but it was the emotional latter parts of the show that really cemented her in my mind. Spoiler for Evangelion spoiler-ish content:
As I stare up at my wall-scroll of Asuka at this moment, I still feel some of that lingering affection. Whatever happens in the Rebuild movies, I'll always have those memories of the original Asuka. And those are some of my best memories in anime, and a formative experience in its own right.
