I recently watched all 8 episodes, and I think Soul Eater is a good show, it's definitely one of the more random out there.
A possibility of becoming a weapon, which also explains Maka not being a weapon, is that it's in the blood but not everyone of the line becomes a weapon so it lies dormant until somewhere along the line you get a weapon. This means they are just human, it also explains why weapons grow older (Stein mentioned it) and why Shinigami has Spirit, instead of an older weapon.

Here's my theory as someone who only watched the anime, instead of genes, blood or having a body that can transform, its the soul instead.
Tsubaki mentioned Mifune as a 'Blade soul'. I believe its your soul that determines whether you can become a weapon. Like Soul probably has a 'Scythe soul' and the sisters' 'Gun souls'.
