So, um, would I be lost if I go into this series blindly (i.e not knowing anything about the Fate series in general outside of the brief summary/premise for Fate/Zero)? If so, how lost would I be or is it easy to pick up?

I think it will be possible to understand it. Especially since I suspect given the massive hype surrounding this show, they'll make it so that those who haven't seen FsN can understand it.

Actually, now that I think about it, what exactly is going to be spoiled for those people aside from:

IMO you should be fine just watching fate zero, you get a more background story if you play the Visual Novel or watch the anime... though you might be disappointed with the anime cause it was poorly made....

Kaiba pretty much nailed it as far as spoilers go. I think watching fate/zero without having any background knowledge at all might be kind of cool, since unlike anyone who has seen any FSN you wont know what the ending is going to be from episode one xP

There's other stuff that I think it would spoiler in FSN, actually. Anything involving the Matou family, certainly, and anything involving Kotomine. However, I think it should be mostly understandable to people who have not played the game, since it's a prequel.

Except it'd ruin part of the fun of Fate/stay night. I say watch Fate/stay night and then Fate/Zero.

Go in order of release, not chronological order.

But the FSN anime was mediocre, and watching it might make one think that Fate/Zero will be mediocre as well.

I say play the visual novel before watching F/Z, or just go straight to Fate/Zero. The first episode is supposed to be a one-hour special, so one can judge whether they would like this series or not by the long introduction material.
