You can start with Shiki, a very well done horror anime. Absolutely serious and mature.

Also Death Note, just in case you've missed it.

Lastly, Ergo Proxy if you enjoy deep psychology/philosophy in a post-apocalyptic scenario.

I'll toss the recent House of Five Leaves into the mix. Other choices with adults include Bartender, Hataraki Man, Bakeneko/Mononoke, Mouryou no Hako, Oh! Edo Rocket, Aoi Bungaku, Twelve Kingdoms, Saiunkoku Monogatari, Nodame Cantabile, Yakushiji Ryoko no Kaiki Jikenbo, Kuragehime, Baccano!, Welcome to the NHK!, REC, and Claymore. Those cover a wide range of genres.

There are a number of shows with children as protagonists that are not all that "childish." Shows like Seirei no Moribito, Kemono no Sou-ja Erin, Dennou Coil, Noein, and Kure-nai all come to mind.

That's at least a start!

I recommend Kara no Kyoukai. It's a series of seven films, each about murders that are taking place through supernatural means. The protagonist is Ryougi Shiki, a girl that has been raised in a family of demon hunters and has the ability to see the death of everything through her Mystic Eyes of Death Perception. She also has an almost insatable urge to kill. It also has some amazing art and animation.

I suspect by "girl in the box" you mean Mouryou no Hako? I listed it above.

The only way to be absolutely certain that an anime is not childish or girlish, you can look for that 18+ label

OK, seriously now, I will agree with a lot of the suggestions above, but be aware that many of those might not be childish, but are IMHO dead boring and/or utterly stupid.

Higurashi No Naku Koro Ni-
At first the anime seems like your typical cutesy moe style anime, but the plot takes a drastic turn for the worse as a murder occurs, shortly followed by the mental derangement of one of the characters as they turn on the others.

Umineko No Naku Koro Ni-
Several members of the Ushiromiya family arrive on a desolate island, each one eager to obtain the rumored inheritance of the family head Kinzo, the rumors circulating around that a large hidden stash of gold left by the witch Beatrice lurks on the island. Things reach a climax as one of the members is found dead. The members of the family begin investigating who did the crime or if it really was the work of a witch.

Jigoku Shoujo-
There exists a rumor about a site called Jigoku Tsuushin, that is accessible at midnight only with the bitter thought of revenge. Upon entering the name of the person you most despise, Jigoku Shoujo will appear before you, bestowing you a small straw doll with a red string. The string is pulled and the person you hate will disappear from the world, vanishing without a trace. However, compensation must be paid, as you too will fall into the depths of Hell upon your death.
