By doing GIF, usually the quality of the sig will drop quite a lot. It should be noted that since GIF's colour bitrate for PS is only 256 as the maximum, there would be a lot of colours that don't blend well-making the sig, well, low-quality. So I don't suggest using GIF extension in making sig unless you are making animation sig OR there is no way to have the signature have it's best condition with JPEG without violating the file size rule. Personally, I think it's better to go for fake-transparency than GIF when the colours have a lot of gradients and colours

To Mion, if you want to do pop up sig, you should try slice tools (For Adobe PS CS 2 lower, it's only in Imageready I think). With slice tools, you could control the non-pop out image to be JPEG while having the pop out images as PNG-24 (which also support transparency, but much, much better than GIF extension since it retains all the colours bits). This will enable you to sneak out the sigs in transparency with low acceptable file size rate.
