At least three major anime publications (all of them with pretty high credentials like the Dragon magazine which is ALL about Kanzaka/Slayers stuff) are on it, so it is pretty much a done deal at this point. That and Megumi herself has hinted it for a while too.
Also, the news basically states an original story, so I'm hoping it is some sort of a hybrid of the official novels and original material (like how some of the first three seasons were).
As for the old cast, I think that is basically a done deal. Original Slayers material + Megumi = all the rest including at least Gourry, Zelgadis and Amelia.
I'm only worried about Amelia's seiyuu returning since she wasn't terribly active in recent years. Compared to the crazy amount of material the Gourry and Zelgadis seiyuus have been involved in.
