Yeah. I think Yamamoto knew an illusion wasn't being used because he could feel Aizen's own reiatsu coursing through the blade. Having said that, Yama-jii probably didn't know that he might've also stopped Aizen from conjuring up illusions on him even if he(Aizen) wanted to.
