Yes, Nabatame Hitomi and Itou Shizuka are great friends IRL, and they even got "married" in a drama CD way back in 2008. Here's the link:
Sadly, Itou Shizuka divorced her real-life husband recently, but to my knowledge, Nabatame Hitomi is still married to her real-life husband.

Even when talking about the 4th wall itself later in the episode, they don't cross it. They come close, but always make the attacks by inference. Usually through the crew discussions on how to proceed, or a line from Kotori.
