Well, they are interested in the Grail and want to kill ALL the masters and servants. But for most of the time they just having "fun" by killing people which they personally consider "evil".

Reika and Jack are still the same. Is implied that the events of the original "Chapter 1" still somehow occurred. Just in the book are not described in details. All we know is that Black Assassin (Jack) was summoned in Tokyo due to "certain circumstances". And now Fiore wants to kill Jack because she killed magus from Yggdmillennia which had summon her.
