Yes and no. They already have in a sense. They have boyfriends but at the same time it's implied that they chose them because they remind them of Koyomi. It could just be a part of the joke but all three of them say repeatly that if they weren't siblings they would have fallen for each other.

Koyomi have been doing some weird stuff with Karen ever since the toothbrush so take it for what it is.

That is both reassuring and somewhat uncomfortable for me. Funny how I'm okay with everything except some part of this. It might be because I have sisters.

However supposedly Tsukihi started dating her boyfriend because he told her that there is no point in calling someone you love oniichan.

They were mentioned in Nise 1 but never makes an appearance. It's kinda hard to say if they are in love. Both of them were planning on giving their boyfriends their first kiss but "someone" decided to beat them to it. Also Tsukihi mentioned that Karen and her boyfriend's relationship have been rocky ever since Nise 1. Their boyfriends also remind them of Koyomi and they did indeed started dating them in order to make him jealous. Tsukihi however also mentions that the reason she started dating was because her boyfriend told her that there is no point in calling the person you love onii-chan.

In conclusion, Karen and Tsukihi are in love with Koyomi but have boyfriends at the same time.

The Araragi children was definitely related seeing how all three of them are dating someone but in love with other people as well.

From episode 7, turns out the Fire Bee is just an illusion after all, so can someone elaborate what sort of method Shinobu applied to diagnose that Karen was stung by a Fire Bee? Her method doesn't seems to be reliable.

She diagnosed her simply by looking at her symptoms. It wasn't stated in episode 7 but Araragi guessed that since Shinobu didn't really wanted to listen to Oshino and because there was too much to remember, she probably forgot that the Bee wasn't for real.
