Really, what I'm looking for is probably an anime version of 1 Litre of Tears. An extraordinary drama that actually made me cry and probably will only be the only drama I'll cry at.
So is there an anime that's similar to 1 Litre of Tears? Or they can be an anime that contain most of the criteria in the topic title: funny, life learning, sad, and bittersweet.
I've seen:
All of Makoto Shinkai's films (even though not funny, love his works)
Welcome to the NHK (fits all the criteria)
Clannad (sad but not tears invoking)
Haibane Renmei(had all the four that I'm looking for. Had it few funny moments, bittersweet, life learning, and sad)

Stein's Gate (great anime. Had all four)
Ano hi mita hana wo bokutachi wa mada shiranai (great anime)
Grave of the Fireflies
Time of Eve
Kimi ga Nozomu Eien (wasn't that great, but I still liked it)
Honey and Clover
Honey and Clover II (great anime again, had all four)
The Girl Who Leapt Through Time
Toradora
School Rumble (wasn't sad, but I love this anime.)

H2O : Footprints in the Sand centers about a orphaned blind boy who enters middle school. He meet 3 three girls who also had sad pasts and he must struggles around the tragedy towards his village. It's bittersweet but it lacks comedy.

True Tears is a romantic drama about a guy who currently lives with his girlfriend until he meet a girl who gives him something unexpected. Again, it has bittersweet moments.

The same goes for Hanbun no Tsuki ga Noboru Sora, ef -a fairy tale of the two- and Angel Beats! (the first and third has both funny and bittersweet moments).

Air - It's about a young traveler looking for a winged being based on a legend his mother told him while he was a child. He meets three girls who might have a connection to the legend and learns that each girl is dealing with sadness and pain in their own way, and resolves to solve them. A beautiful heartwarming and tragic anime, totally one of my top three.

Puella Magi Madoka Magica - It's about a girl named Madoka who lived an ordinary life until she had life changing encounter with a girl named Homura Akemi and a wish granting creature named Kyubey. Starts off heartwarming but the pain and tragedy starts to pick up immediately after. My favorite anime to date.

Kowarekake no Orgel - A young ex musician finds a discarded robot girl and they start living together. While living together the robot girl realizes that her new owner/friend is struggling with personal issues. It's a single 30 minute OAV, but it's one of the most beautiful stories I've ever watched. I totally recommend it.

I'll suggest Kemono no Sou-ja Erin. It has numerous emotional highs and lows as it tells the story of a young girl who is capable of communicating with mystical beasts. There's a lot more going on than just that premise, too. Don't be put off by the "child-like" style of illustration either. (I can only link to the Crunchyroll stream since it's licensed, but the fansubs are better translated.)

For a more mature show, I recommend Bartender.

I'll second Usagi Drop and suggest another noitaminA show, Tokyo Magnitude 8.0. Dennou Coil might be another possibility; it's a show everyone should watch regardless of their interests. There's also some of what you're looking for in Cross Game.

Haven't really encountered anything like 1 Litre of Tears, but maybe Winter Sonata is worth a try. I haven't seen it personally.

Saikano was another good one ... very sad though.

As for Fruits Basket ... dunno if you are a manga reader but the manga was 10x better imo. It'd be worth it to read it if you like the anime imo.

Neither my daughter nor I found the anime version of Winter Sonata compelling enough to continue past an episode or two. I hear the live-action K-drama version is better, but I haven't seen it.

I personally find the K-drama version to be much better. The anime version lacks the impact of the characters' emotion that is very visible in the drama version. Just my two cents.

Then Ryo will watch the K-drama version instead.

I second Madoka Magica too. First three episodes look sweet and a bit fluffy, but after that lot of pain and tragedy will occur as the story progresses.

He prefers the newer animation, but Grave of the Fireflies is the only old drama movie he have seen.

One anime that immediately came to mind when I read the title of this thread is Brigadoon. Don't be fooled by how silly and cartoony this looks.

In the beginning, the characters live a happy, although occasionally dangerous life together. However, many complications arise and as the story progresses, the characters are put through hell.

The dark side of humanity is explored, the characters are hit by painful and traumatic moments, and people die. But despite all that, the love (romantic and platonic) that the characters feel for each other keeps them going and motivates their actions.

Brigadoon is a very powerful anime with lots of happy and funny moments contrasting with heartrending drama.

Rainbow: Nisha Rokubou no Shichinin is a great show. Can't say it has much comedy but all other requirements are met well.
