Thing i don't see on Esdese is she straightly kills them even they possess strong will unlike from other manga such as one piece Mihawk want to see how Zoro has grown to be a worthy opponent in the future.

After all Tatsumi has to escape many times from Esdese while Suu remembered by Esdese as a warrior not a teigu before Suu's demise. Frankly enough Esdese's current worthy opponent is only Tatsumi.

I am debating on whether or not to read this. I know that there is rape in the series. Is it similar to the sexual assault moments in "Seikon no Qwaser" (the soma-sucking moments and the Hana/Katja moments)?

I would say it's pretty different. It's usually off-screen. It's also to be taken more seriously as it usually comes along with torture and murder.

It doesn't happen that often though.

No, that's more fanservice. This is more blood boiling type of assault.

Anyway, I really hope the author doesn't kill off Wave and Kurome, after all that it would be such a distasteful move.
