Have you watched Moyashimon? Haruka is a pretty good rendition of a tsundere.

Love Hina - My fave
Honey and Clover - I don't remember what parts were funny, but its been a long time since i've seen this one. It does have humor tho.
Kamisama Kazoku - This one is pretty good. the love story alone is hilarious
Nodame Cantabile - GREAT anime. Very Funny
Fruits Basket - Funny, not hilarious, but not supposed to be.
Ouran High School Host Club - Great Anime all around. pretty funny, nice bit of romance... kinda

(Random Pick of the day) Ebichu This is Ecchi (not hardcore by any means) But is Absolutely HILARIOUS! Slight romance, if u want to call it that. still funny enough to recommend.

Kagami =/= tsundere.
Or neo for that matter.

Definitely try out Love Hina, Naru is one of the reasons tsunderekkos are popular nowadays.
Zero no Tsukaima has a loli-tsunderekko, it's a bit of a mix with action/adventure/ecchi, though.
Hayate no Gotoku came out within the last year, so you've probably seen it, but if not, Nagi's one of the better recent neo-tsundere characters.
Full Metal Panic! has Kaname, who is a tsunderekko filled with win. It has a bit of mecha action mixed in, but there's a lot of comedy there.

Ah comedy, you got to love it

Basically, I recommend the following:

Hayate no Gotuku, this is one of my favourite, its pure randomness will just throw you off (though I don't think its randomness can rival Lucky Star's... )

Inukami! is another that gave me a HUGE laugh. It is one of the funniest around I've got to say. But one thing though, try survive the first two episodes of Inukami!. I know it is VERY disturbing, but it gets less (or does it ?)

Love Hina is another great romance comedy if you are interested. If this is your type, I suppose I could also recommend Ah! My Goddess, it is quite a sweet anime, not as much laugh as the first two but I still enjoyed it.

Clearly, you've yet to watch a SHAFT series.

Pani Poni Dash is pretty much as random as it gets.
Sayonara Zetsubou Sensei is one of the better recent comedies, but you've probably seen it.

No tsunderekkos in either, though.

And no, Inukami! is disturbing from start to finish, but that's what makes it great.
